Submitting Samples:
1. Samples are to be provided in non-stick nuclease free vials (Life technologies cat # AM12350) in 5ul sterile PBS
(Life Technologies cat # 14040-141) for cells and nuclease free water (Life Technologies cat # AM9938) for nucleic
acids
2. Customers should prepare two sets of labels for each sample, one should be applied to the sample tube and the other
provided to us for labeling the amplified/labeled product
3. Samples should be shipped on dry ice along with details description of each sample and labeling pattern
Product Delivery:
1. Amplified nucleic acids are shipped in nuclease free water
2. Fluorescent labeled cDNA, miRNA, gDNA or probes shipped freeze-dried sealed under dry nitrogen
3. Biotin labeled cRNA/miRNA are shipped in nuclease free water
5. The spare label provided by the customer is applied on the service products
6. Service samples shipped on dry ice along with shipment label and quality analysis
7. PCR primers and standards are shipped as freeze-dried aliquots in room temperature
Product Deliverables:
1. Amplified nucleic acids:
a. Quantification by spectrophotometry (OD 260)
b. Size estimation by electrophoresis (gel electrophoresis images)
c. Samples normalized to 100ng/ul in Nuclease Free Water
2. Fluorescent labeled cDNA/gDNA:
a. Quantification by spectrophotometry (OD 260)
b. Dye incorporation quantified by spectrophotometry
c. Size estimation by electrophoresis (gel electrophoresis images)
d. Samples freeze-dried in 2ug aliquots under dry nitrogen
3. Biotinylated cRNA/miRNA for Affymetrix chips:
a. Quantification by spectrophotometry (OD 260)
b. Size estimation by electrophoresis (gel electrophoresis images)
c. Samples normalized to 100ng/ul in Nuclease Free Water
4. Fluorescent labeled miRNA:
a. Quantification by spectrophotometry (OD 260)
b. Dye incorporation quantified by spectrophotometry
c. Size estimation by electrophoresis (gel electrophoresis images)
d. Samples freeze-dried in 2ug aliquots under dry nitrogen
5. Primers and standards for mRNA quantification
a. Mixed forward and reverse primers: (2.5 nmol lyophilized (sufficient for 250 reactions)
b. Gene Specific copy number standard: 100 million copies (double-stranded DNA) lyophilized
6. Primers and standard for miRNA quantification
a. 200 pmoles dTVN primer with an overhang (20 RT reactions)
b. Mixed forward and reverse primers: (2 nmol lyophilized (sufficient for 200 reactions)
c. Gene Specific copy number standard: 100 million copies (double-stranded DNA) lyophilized
7. Probes for in situ hybridization
a. Upto 12 probes per mRNA and single probe per miRNA
b. Upto 15-20 flurochromes per probe
c. 2 nmoles of labeled oligos shipped freeze-dried sealed under dry nitrogen
